摘要:采用正交試驗設計法,以綠原酸為對照,分析物液比、提取溫度、提取時間、乙醇體積分數4個因子對通關藤(Marsdenia tenacissima)中總酚酸提取率的影響。結果表明,影響提取率的主次因素為乙醇體積分數、提取時間、料液比、提取溫度,最佳提取條件為乙醇體積分數70%,提取時間1.0 h,料液比1∶16(m/V,g∶mL),提取溫度50 ℃。

Technology of Extracting Total Phenolic Acid from Marsdenia tenacissima

Abstract: The orthogonal design was used to analysis optimize extraction process of total phenolic acid from Marsdenia tenacissima. Four factors including the material ratio, extracting time, extraction temperature and mass fraction of ethanol were sudied using chlorogenic acid as control. The results showed that the order of the four factors affecting extraction efficiency was as follows: mass fraction of ethanol > extraction time > material liquid ratio >extraction temperature. The optimal extraction process of chlorogenic acid was mass fraction of ethanol 70%, extracting time 1 h, material liquid ratio 1∶16, extraction temperature 50 ℃.

通關藤(Marsdenia tenacissima)為蘿摩科牛奶菜屬植物,別名烏骨藤、大苦藤、扁藤、下奶藤等。主要分布于我國云南、廣西、貴州、四川等地及越南、老撾、緬甸等東南亞國家,具有清熱解毒、祛痰平喘、散結止痛的作用,用于氣管炎、支氣管哮喘等病的治療[1]。曾收載于《云南省藥品標準》和《中國藥典》,具有免疫調節、保肝利尿、敗毒抗癌等作用,對胃癌、肝癌等多種腫瘤具有確切的抗腫瘤療效[2,3]。通關藤是一種重要的中藥材,以通關藤為單味藥制成注射液、糖漿、口服液、片劑、膠囊等多種劑型,含有甾體酯苷、生物堿、有機酸、多糖、樹脂及色素等多種化學成分[4]。通關藤中綠原酸含量較高,為有效成分之一[5], 綠原酸有抗誘變、抗菌、抗病毒等作用,而多酚類物質具有殺死癌細胞,使人肝癌細胞、胃癌細胞和肺癌細胞向正常細胞逆轉的作用[6]。對綠原酸的提取方法進行研究,可為制藥企業提供參考依據。
