中藥及復方水提物對DPPH·和·OH的清除作用

摘要:采用分光光度法分析測定了具有抗仔豬腹瀉作用的12種中藥及5個復方的水提物對1,1-二苯基-2-苦基肼自由基(DPPH·)和羥基自由基(·OH)的清除能力,并對其多糖含量進行了測定。結果表明,12種中藥及5個復方對DPPH·和·OH均有一定程度的清除作用。濃度在0.2~1.0 mg/mL范圍內,秦皮、白頭翁、黃連、黃柏以及復方2、4、5對DPPH·清除作用較好,在高濃度時與維生素C對DPPH·的清除作用相當。除復方3外,中藥復方在高濃度時對DPPH·的清除率接近或高于其組成中藥,表現出協同效應。濃度在5~25 mg/mL范圍內,白術、防風、魚腥草及復方1和復方3對·OH的清除作用較好,復方1和復方3表現出對·OH清除的協同作用。中藥及其復方多糖含量與DPPH·和·OH清除作用不存在明顯的相關性。

Scavenging Effect of Aqueous Extracts from Some Anti-diarrhea Chinese Herbs and Compounds on DPPH· and ·OH

Abstract: Using spectrophotometric and phenol-sulfuric acid method, the scavenging effect on DPPH and hydroxyl radicals, and the polysaccharide contents in the aqueous extracts from 12 Chinese herbs and 5 compounds with anti-diarrhea effects were investigated. The results showed that all 12 Chinese herbs and 5 compounds had scavenging effect on DPPH· and ·OH with different degree. With the concentration of 0.2 to 1.0 mg/mL, the Cortex fraxini, Radix pulsatillae, Rhizoma coptidis and Cortex phellodendri and the compounds 2, 4 and 5 had high scavenging effects on DPPH·. The scavenging effect was close to that of vitamin C at 0.8 mg/mL and 1.0 mg/mL. Except the compounds 3, other compounds had synergistic effect and showed higher scavenging effect on DPPH than that of its component herbs with tested concentration. Under the concentration of 5 mg/mL to 25 mg/mL, the extracts from Rhizoma atractylodis, Radix saposhnikoviae and Houttuynia cordata and compounds 1 and 3 had good scavenging effect on hydroxyl radicals. Similarly, the compounds 1 and 3 showed synergistic effect. There was no significant correlation between the polysaccharide content of different herbs, compounds and the scavenging effect on DPPH· and ·OH.

早期斷奶仔豬因消化器官發育不充分、消化酶分泌不足、免疫力低下等極易帶來嚴重的應激反應,導致腸道系統紊亂及腹瀉[1]。正常情況下,機體內自由基的產生與清除處于動態平衡,維持著機體的正常生理機能,但當仔豬在遭受患病、應激或者日糧中營養缺乏等情況下,機體代謝出現異常,自由基將過量產生。自由基活性極高,攻擊性很強,導致機體發生一系列變化,致使動物病理狀況加重[2,3]。中國全年仔豬平均腹瀉發病率為46.5%[4],因腹瀉死亡的仔豬占仔豬總數的39.8%[5],給養豬業帶來巨大的經濟損失。……
