摘要:為探討滅多威對家蠅胚胎細胞系MDEC-07114細胞周期及細胞周期蛋白Cyclin-D1的影響,用流式細胞儀檢測不同濃度滅多威對MDEC-07114細胞周期的影響,應用免疫組化S-P法(Streptavidin-perosidase)檢測細胞周期蛋白Cyclin-D1表達。結果表明滅多威作用于MDEC-07114后,各濃度組G0/G1期升高,S+G2/M期細胞占比及PI降低;從滅多威1 mg/mL處理組開始Cyclin-D1表達出現降低,且隨濃度的增加降幅增大??梢姕缍嗤梢酝ㄟ^下調Cyclin-D1使細胞周期阻滯而抑制細胞生長。

Effects of Methomyl on Cell Cycle and Cyclin-D1 of MDEC-07114 Cell

Abstract: To investigate the effects of Methomyl on cell cycle and cyclin-D1 of MDEC-07114 Cell, experiments were conducted to measure the effects on cell cycle with flow cytometry and expression of cyclin-D with immunohistochemical S-P method. For the MDEC-07114 cell under Methomyl, the percentage of G0/G1 was increased while the percentage of S+G2/M and the cell proliferation index were obviously reduced at the all concentration-groups. From the 1mg/mL groups of Methomyl, the expression of cyclin-D1 was reduced with the increase of the concentration.Therefore, the growth of MDEC-07114 may be inhibited by reducing expression of cyclin-D1 to block cell cycle.

MDEC-07114是遵義醫學院寄生蟲學課題組自建的家蠅胚胎細胞系,滅多威為氨基甲酸酯類殺蟲劑,對昆蟲卵有殺傷作用,對家蠅也有毒殺作用,前期研究表明滅多威對體外培養的MDEC-07114細胞有抑殺作用[1]。家蠅抗藥性問題日趨嚴重,尋找新的殺蟲劑成為必然。昆蟲細胞凋亡的研究是探討昆蟲殺蟲劑作用機制、尋找昆蟲防治新途徑的探索和嘗試。細胞凋亡與細胞周期阻滯有關[2-4]。本試驗研究滅多威對MDEC-07114的細胞周期及細胞周期調控蛋白的影響,為新的昆蟲防治方法提供思路。
1 材料與方法
1.1 材料
1.1.1 供試細胞 家蠅胚胎細胞系MDEC-07114,由遵義醫學院寄生蟲學教研室提供。
1.1.2 滅多威溶液 滅多威[含量99.9%,批號BW(E)060546] 購自北京恒元啟天化工技術研究院,丙酮(分析純)購自成都化學試劑廠。先用無血M3培養基溶解丙酮,配制成1%丙酮溶液,抽濾,4 ℃保存。然后用1%丙酮溶液溶解滅多威,混勻抽濾配成20 mg/mL滅多威母液,4 ℃保存。使用時用1%丙酮溶液稀釋成不同濃度供試驗用。
1.1.3 碘化丙啶溶液 將碘化丙啶(美國Sigma公司生產)溶于PBS中,終濃度為100 mg/mL,含RNase 100 μg/mL,4 ℃避光保存?!?br>
