β-環糊精催化合成N-苯基鄰苯二甲酰亞胺

以β-環糊精為催化劑,苯胺和鄰苯二甲酸酐為原料,在水溶劑中合成N-苯基鄰苯二甲酰亞胺,考察了反應時間,催化劑用量,胺酐比,溶劑用量等因素對產物收率的影響。結果表明,β-環糊精有著很好的催化活性,當反應時間為5h,胺酐比為1.3∶1(mol),β-環糊精濃度1%(以溶劑質量計)時,在30mL加入少量鹽酸的水溶劑中,收率可達到88.7%,且催化劑重復使用性好。

Abstract:N-phenylphthalimide was synthesized with using aniline and phthalic anhydride as raw materials and β-cyclodextrin as catalyst in the presence of hydro-solvent.The effects of reaction time,catalyst amount,ratio of aniline to phthalic anhydride and solvent amount on the production yield were investigated.The results showed that the β-cyclodextrin had excellent catalytic activity,and the optimum reaction conditions were as follows:the molar ratio of aniline to phthalic anhydride was 1:1.3,the amount of hydro-solvent with a small dosage of hydrochloric acid was 30mL,the reaction time was 5h and the amount of catalyst was 1%(mass)of solvent;the yield of product was up to 88.7%,and the stability of reused catalyst was excellent.

N-苯基鄰苯二甲酰亞胺是白色結晶性粉末,通常由苯胺和鄰苯二甲酸酐制取,是制備吲哚洛芬和2,3,4,5-四氟苯甲酸的關鍵中間體,在生物、醫藥及生命科學領域有著廣泛應用[1]。而2,3,4,5-四氟苯甲酸是洛美沙星、氧氟沙星、左旋氧氟沙星、斯帕沙星的起始原料,故其中間體N-苯基鄰苯二甲酰亞胺的合成越來越引起了人們的重視[2]。
目前,N-苯基鄰苯二甲酰亞胺主要以鄰苯二甲酸酐與苯胺為原料在有機溶劑和無機鹽反應體系中合成,但是普遍存在著有機溶劑毒性大,揮發性強,反應催化劑難于分離,純化過程過于復雜等一系列問題[3]。
本文主要研究以苯胺和鄰苯二甲酸酐為原料,以水作為溶劑,采用膠束催化法以β-環糊精為催化劑合成N-苯基鄰苯二甲酰亞胺。產物和催化劑易分離,沒有難于除去的雜質存在,不但大大提高了產品的純度,而且秉持綠色化學理念,環保無污染。……
