山桔葉水提物急性毒性試驗

Acute Toxicity Experiment of Glycosmis parviflorae（Sims）Kurz Water Extract

Abstract：ObjectiveToevaluate theacutetoxicityof Glycosmis paruflorae（Sims）Kurz water extract inmice.Methods The maximum dosage（MAD）wasused inthe acute toxicityexperiment on mice.40 KMmice，2O malesand2O females，were selected andrandomlydividedintotwogroups with2Omiceineachgroup.Thetreat groupwasgiventhewaterextractofGlycosmisparviflore at a volume of 40mL/kg by gavage，and the control group was given equivalent pure water， observed and recorded general condition of mice（toxicreaction，body weightanddeath）within14days.After14days，theserum wastakentodetectthebloodbiochemical indexes，observedthepathologicalchangesinthemainorgansadcaleulatedtheorganindex.Inadition，histopathologicalchangs inthe organ wereanalyzedby hematoxylinandeosin method.ResultsThere werenoobvious toxic symptoms inthetreat group，and nosignificantdiferenceintebdyweights，matologicalandochemicalparametersaswellasistopatologyinorganshncompared with thecontrol group.Conclusion The MAD of water extractof Glycosmis paruflorae（（Sims）Kurz for mice was 218g/kg ， which was 872 times of the recommended dosage for adults （ 60kg ）.Indicatedt hat Glycosmis parviflorae（Sims）Kurz was high safety.

山桔葉為蕓香科山小橘屬小花山小橘Glycosmisparviflorae（Sims）Kurz的干燥葉，別名野沙柑、酒餅木、山油甘等，具有止咳、化痰、消積、破瘀消腫的功效，在廣西、廣東、福建、臺灣等均有分布[1-2]。民間常用于治療感冒咳嗽、食積脹痛等。化學研究[3-7]表明，山桔葉主要含萜類、甾體類、黃酮苷類、脂肪酸類化合物。而其藥理作用報道較少，僅見抗炎、鎮痛相關報道[8-9]。基于此，本研究將初步探討山桔葉水提物的急性毒性，為臨床上合理、安全使用山桔葉提供參考。

1材料與方法

1.1材料谷草轉氨酶（aspartate amino transferase，GOT/AST，批號：20220621），谷丙轉氨酶（alanineamino transferase，GPT/ALT，批號：20220615），超氧化物歧化酶（auperooxidedismutase，SOD，批號：20220624），谷胱甘肽過氧化物酶（glutathioneperoxi-dase，GSH-PX，批號：20220704）。

山桔葉采自廣西南寧市伊嶺巖，經廣西中醫藥大學中藥鑒定教研室蔡毅教授鑒定為蕓香科植物小花山小橘Glycosmisparviflorae（Sims）Kurz的干燥葉。

1.2儀器BSA224S型電子天平（賽多利斯科學儀器有限公司）；HWS－26型電熱恒溫水浴鍋（上海一恒科學儀器有限公司）；TGL-16G型高速離心機（上海安亭科學儀器廠）； M200Pro 型連續波長檢測儀（TECAN帝肯）。

1.3動物KM種小鼠，體重（ 20±2 ）g，雌雄各半，由湖南斯萊克景達實驗動物有限公司提供，動物許可證號：SCXK（湘） 2019-0004 。飼養環境：室溫 20～30^°C ，相對濕度 60%～80% 。廣西中醫藥大學實驗動物中心資質號：SYXK桂 2019-0001 。

1.4方法

1.4.1山桔葉水提液制備取干燥的山桔葉藥材1500g ，加12倍量水煎煮3次，提取時間依次為2h、 ¹h 、1h，提取液濾過合并，減壓濃縮至以灌胃針能通過的最大濃度（ 5.45g 生藥/ ΔmL ）的山桔葉水提濃縮液。

1.4.2小鼠急性毒性實驗預實驗取20只KM種小鼠，體重（ 20±2 ） g ，雌雄各半，隨機分為空白對照組及給藥組，每組……
