摘 要 ???:為探究雌雄異株植物響應重金屬脅迫的性別差異,本研究利用超高效液相色譜結合四級桿飛行時間質譜儀(UPLC-Q-TOF-MS)測定了鎘(Cd)脅迫下雌雄地錢( Marchantia polymorpha )中的代謝產物,分析了與不同性別相關的差異代謝物和代謝通路.結果顯示,未受鎘脅迫時,雌性地錢中類黃酮生物合成、嘌呤代謝和玉米素生物合成代謝水平高于雄性,芹菜素、木犀草素、腺嘌呤和腺苷含量顯著高于雄性.受鎘脅迫后,雌性地錢中類黃酮生物合成、嘌呤代謝和玉米素生物合成代謝水平下降,芹菜素、木犀草素、矢車菊素-3-O-半乳糖苷、腺嘌呤和腺苷含量顯著下降.而雄性地錢中上述三條通路的代謝水平上升,刺槐苷、槲皮素、腺苷一磷酸和肌苷的含量顯著上調.研究表明,地錢響應鎘脅迫的代謝變化出現了性別差異.其中,雌性地錢降低了玉米素和遺傳物質的合成水平,誘導谷胱甘肽的合成.雄性地錢上調了玉米素合成,緩解葉綠素的降解,同時上調類黃酮生物合成途徑中的槲皮素和刺槐苷等具有抗氧化活性的次生代謝產物.

Study on gender response difference of ?Marchantia polymorpha ??metabolome under cadmium stress based on LC-MS technique

In order to explore the gender differences of dioecious plants in response to heavy metal stress, the metabolites of ?Marchantia polymorpha ?under cadmium (Cd) stress were determined by ultra-performance liquid chromatography combined with quadrupole time-of-flight mass spectrometry ( UPLC-Q-TOF-MS ), and the differential metabolites and metabolic pathways related to different genders were analyzed. The results showed that, without cadmium stress, the levels of flavonoid biosynthesis, purine metabolism and zeatin biosynthesis in females were higher than those in males with significantly higher contents of apigenin, luteolin, adenine and adenosine. After cadmium stress, the levels of flavonoid biosynthesis, purine metabolism and zeatin biosynthesis in females were decreased with the significant decrease of ?apigenin, luteolin, cyanidin-3-O-galactoside, adenine and adenosine. The metabolic levels of the above three pathways increased in males with the significant increase of differential metabolites kaempferol-3-O-robinoside-7-O-rhamnoside, quercetin, adenosine monophosphate and inosine. Studies have shown gender differences in metabolic changes of ?M. polymorpha ?in response to cadmium stress. Among them, female ?M. polymorpha ?reduced the synthesis level of zeatin and genetic material, and induced the synthesis of glutathione. Male ?M. polymorpha ?increased zeatin synthesis, alleviated chlorophyll degradation, and up-regulated secondary metabolites with antioxidant activity such as quercetin and kaempferol-3-O-robinoside-7-O-rhamnoside in the flavonoid biosynthesis pathway.
