為了研究擬南芥脫落酸(Abscisic acid,ABA)受體的翻譯后修飾,本研究以能夠磷酸化ABA受體的激酶CARK5作為研究對象,通過構建了轉基因株系來檢測CARK5在ABA信號途徑中的功能.結果顯示: CARK5能夠促進ABA介導的抑制種子萌發、幼苗的形態建成以及主根的生長,但是過表達激酶失活型CARK5m(CARK5N250A)則跟突變體cark5表型類似.擬南芥中過表達CARK5增強植株抗旱性;ABA處理后,ABA響應標記基因RAB18表達量增加.這些結果說明,CARK5通過可以磷酸化ABA受體,正調控ABA信號通路.

Response of CARK5 kinase to ABA signaling in Arabidopsis

In order to explore the post-translational modification of Arabidopsis abscisic acid (ABA) receptor, this study constructed overexpressing CARK5 and kinase-dead CARK5m (CARK5N250A) in the wild type and T-DNA mutant cark5 plants by genetic and physiological assays, to analyze the functions of CARK5 in ABA signaling. The results showed that CARK5 can promote ABA-mediated inhibition of seed germination, seedling cotyledon and primary root elongation, but the overexpression kinase-inactive CARK5m (CARK5N250A) has a similar phenotype to the mutant cark5. In addition, overexpressing CARK5 enhanced plant drought resistance and ABA-marked gene RAB18 expression. Taken together, these data suggest that CARK5 positively regulates the ABA signaling pathway by phosphorylation of ABA receptors.

1 引 言
植物能夠通過捕捉環境信號,然后激活植物激素介導的多種內源途徑,進而影響植物從種子成熟、種子萌發到開花結果以及響應生物或者非生物脅迫等各種生命活動[1,2].其中脫落酸(Abscisicacid,ABA)是一種由類胡蘿卜素裂解而成的重要植物激素[3],在植物中具有多種生理功能.ABA通過調控種子的休眠和萌發以及氣孔的開度等生理活動,來應對干旱、高溫、高鹽等環境脅迫[4].擬南芥響應ABA信號通路的過程是:首先ABA受體PYR1/PYLs/RCARs(Regulatory Components of ABA Receptors/Pyrabactin Resistance/PYR like)感知ABA后,能夠抑制磷酸酶PP2C活性,激活激酶SnRK2s,使其磷酸化ABA信號通路的下游信號分子,最終響應ABA信號.ABA受體作為ABA信號通路中的核心成員[5-7],在擬南芥中總共有14個成員[8].在14個成員中除了PYL13以外,其余13個都被證實能夠結合ABA.
