摘要:目的? 建立超高效液相色譜法同時測定鹽膚木中8種成分含量方法,為鹽膚木藥材的質量控制提供依據。方法? 采用ACQUITY UPLC BEH C18色譜柱(2.1 mm×100 mm,1.7 ?m),流動相為乙腈-0.1%甲酸水梯度洗脫,流速0.3 mL/min,柱溫35 ℃,檢測波長275 nm。結果? 沒食子酸、苔黑酚葡萄糖苷、沒食子酸甲酯、3,5-二羥基甲苯、鞣花酸、漆黃素、五沒食子酰葡萄糖和根皮苷的線性范圍分別為23.31~1165.5 ng(r=0.999 2)、27.02~1351.5 ng(r=0.999 1)、6.30~315.0 ng(r=0.999 2)、12.36~618.0 ng(r=0.999 2)、17.14~857.5 ng(r=0.999 6)、9.24~461.5 ng(r=0.999 8)、25.29~1264.5 ng(r=0.999 6)和6.18~309.0 ng(r=0.999 3),平均加樣回收率為96.77%~99.41%(RSD為2.23%~3.24%)。鹽膚木樣品經聚類分析和主成分分析可分為兩類,且沒食子酸、五沒食子酰葡萄糖、沒食子酸甲酯和漆黃素等成分在兩類中的分布存在差異。結論? 本研究建立的方法可為完善鹽膚木藥材的質量標準和加強質量控制提供依據。

Simultaneous Content Determination of Eight Compositions in Rhus Chinensis Mill. by UPLC

Abstract: Objective To establish a UPLC method to simultaneously determine the contents of eight chemical compositions in the root of Rhus chinensis Mill.; To provide references for quality control of Rhus chinensis Mill. Methods The UPLC method was performed on an ACQUITY UPLC BEH C18 column (2.1 mm × 100 mm, 1.7 ?m) with a gradient elution using acetonitrile and 0.1% formic acid. The flow rate was 0.3 mL/min; the column temperature was 35 ℃; the detection wavelength was set at 275 nm. Results Gallic acid, orcinol glucoside, methyl gallate, 3,5-dihydroxytoluene, ellagic acid, fisetin, pentagalloylglucose and phlorizin showed good linear relationship within the range of 23.31–1165.5 ng (r=0.999 2), 27.02–1351.5 ng (r=0.999 1), 6.30–315.0 ng (r=0.999 2), 12.36–618.0 ng (r=0.999 2), 17.14–857.5 ng (r=0.999 6), 9.24–461.5 ng (r=0.999 8), 25.29–1264.5 ng (r=0.999 6), and 6.18–309.0 ng (r=0.999 3), respectively. The overall recoveries were ranged from 96.77% to 99.41% with the RSD range of 2.23%–3.24%. After cluster analysis and principal components analysis, the collected samples of Rhus chinensis Mill. could be divided into two types of patterns, and there are differences in the distribution of compositions in the two types, such as gallic acid, pentagalloylglucose, methyl gallate and fisetin. Conclusion The method can provide a basis for improving the quality standard and quality control of Rhus chinensis Mill.

鹽膚木Rhus chinensis Mill.又稱五倍子樹、五倍柴等,屬漆樹科鹽膚木屬的落葉灌木或喬木,主要分布于福建、廣東、廣西等地,其主要入藥部位為根和莖,有活血化瘀、行血止痛功效,用于治療冠心病、心絞痛、胸悶、憋氣等[1]。現代研究表明,鹽膚木具有改善微循環和心腦缺血、抗血小板凝集、降脂、抗炎、抗人類免疫缺陷病毒等活性,其研究和應用潛力很大[2-4]。目前僅《福建省中藥材標準》[5]和《廣東省中藥材標準》[6]收載鹽膚木,而國內外文獻對鹽膚木藥材質量控制的報道很少。有報道以沒食子酸含……
