摘要:角鯊烯環氧化酶(SQE)催化角鯊烯合成三萜類骨架β-香樹酯前體2,3氧化鯊烯,是熊果酸合成途徑中的重要限速酶。本研究采用RT-PCR 技術從‘喜來藍莓根中克隆得到VcSQE基因,全長序列1 744 bp,序列分析結果表明,其含有1 620 bp開放閱讀框,編碼539個氨基酸;氨基酸同源序列分析表明,藍莓與其它物種SQE蛋白氨基酸序列相似性很高,與山茶科的茶相似性最高,達90.6%,與獼猴桃相似性為89.7%;不同植物SQE蛋白序列分析發現,該類植物SQE含有3個保守結構域,均在VcSQE中存在;熒光定量試驗表明,VcSQE基因在葉中表達量最高,根中表達量最低。

Cloning and Expression Analysis of a SQE Homologous Gene from
Blueberry(Vaccinium corymbosum L.)

Abstract Squalene epoxidase (SQE) catalyzes the synthesis of triterpenoid β-fragrant resin precursor 2,3 squalene. It is an important rate-limiting enzyme in the ursolic acid synthesis pathway. The VcSQE gene was cloned from the Xilai blueberry root by RT-PCR with the full-length sequence as 1 744 bp. The sequence analysis showed that its open reading frame was 1 620 bp,and it encoded 539 amino acids. The amino acid homologous sequence analysis showed that the amino acid sequence of SQE protein in Vaccinium corymbosum L. was very similar to others. The highest similarity was 90.6% with that of tea in Camellia, and the similarity with Actinidia chinensis var. chinensis was 89.7%. Plant SQE had 3 conserved domains, and they were all present in VcSQE. The fluorescence quantitative experiments showed that the VcSQE gene had the highest expression in blueberry leaves and the lowest in roots.

藍莓作為一種高營養和高市場價值的水果,在中國的栽培面積和栽培種類日趨增多,而伴隨藍莓采后加工工藝的研發和優化,市場需求量越來越高[1-5]。藍莓含有水果中普遍存在的營養物質糖、酸、礦物元素等,同時也含有大量尼克酸、黃酮等[6-8]。此外,還含有三萜類物質熊果酸和齊墩果酸[9]。
熊果酸(ursolic acid, UA)是存在于植物中的五環三萜類化合物,以游離酸或皂苷的糖苷配基形式存在。已被證實存在于許多藥用植物中,如枇杷、粳稻、迷迭香、白花蛇舌草,蘋果、梨和李子的蠟層中也發現有熊果酸存在[10]。因其具有一定的生物活性和低毒特性,現已成為許多生命科學領域研究的熱點。已有研究證明熊果酸具有抗腫瘤、保肝、消炎等多種功效,能夠抵抗多種致癌及促癌物[11],并在抗氧化、拮抗微生物、降低血脂、鎮痛、保護心血管、抗動脈粥樣硬化和免疫調節等方面發揮作用[12,13]。……
