摘要?目的:觀察不同電針介入時機對腰多裂肌損傷大鼠多裂肌叉頭蛋白(Foxo1)、肌肉生長抑制素(Myostatin)、成肌分化因子(Myod)蛋白表達的影響。方法:雄性SD大鼠隨機分為空白組、模型組、即刻電針組、24 h電針組、48 h電針組,每組8只。以多裂肌肌注0.5%布比卡因復制腰多裂肌損傷模型。各電針組分別在造模后即刻、24 h、48 h開始電針雙側“委中”“腎俞”,連續干預7 d。以Western blot檢測各組多裂肌Foxo1、Myostatin、Myod蛋白表達水平。結果:與空白組比較,模型組Foxo1、Myod蛋白表達水平顯著升高(P<0.01);與模型組比較,即刻電針組、48 h電針組Foxo1、Myostatin蛋白表達水平降低(P<0.01,P<0.05),24 h電針組Foxo1、Myostatin蛋白表達水平顯著降低(P<0.01),Myod蛋白表達水平顯著升高(P<0.01);與24 h電針組比較,即刻電針組Myostatin蛋白表達水平升高(P<0.05)、Myod蛋白表達水平顯著降低(P<0.01),48 h電針組Foxo1蛋白表達水平顯著升高(P<0.01)、Myod蛋白表達水平顯著降低(P<0.01)。結論:電針促進多裂肌修復的最佳介入時機可能為損傷后24 h。

Effects of Electroacupuncture Intervention Time on the Expression of Foxo1,Myostatin and Myod in Rats with Multifidus Muscle Injury

Abstract?Objective:To observe the effects of different electroacupuncture intervention time on the expression of multifidus muscle fork protein(Foxo1),Myostatin and myoblast differentiation factor(Myod)in rats with lumbar multifidus muscle injury. Methods:Male Sprague-Dawley rats were randomly divided into the blank group(BG),model group(MG),immediate/24 h/48 h electroacupuncture group(0 hG/24 hG/48 hG),with 8 rats in each group. The lumbar multifidus muscle injury model was replicated with 0.5% bupivacaine in the multifidus muscle. Each electroacupuncture group started the electroacupuncture on both sides of the Weizhong(BL40)and Shenshu(BL23)at 0 h/24 h/48 h after modeling,and continuous intervention for 7 days. Western blot was used to detect the expression levels of Foxo1,Myostatin and Myod proteins in multifidus muscles of each group. Results:Compared with the BG,the expression of Foxo1 and Myod proteins in the MG significantly increased(P<0.01); Compared with the MG,the expression of Foxo1 and Myostatin in the 0 hG and 48 hG were decreased(P<0.01,P<0.05). The expression of Foxo1 and Myostatin in the 24 hG significantly decreased(P<0.01),and Myod protein expression was significantly increased(P<0.01). Compared with the 24 hG,the expression of Myostatin protein in 0 hG increased(P<0.05),and the expression of Myod protein in 0 hG significantly decreased(P<0.01); the expression of Foxo1 protein in 48 hG was significantly increased(P<0.01),and the expression of Myod protein was significantly decreased(P<0.01). Conclusion:The best intervention time is 24 h after injury,wich can effectively promote multifidus muscle repairing.

多裂肌是附著面積最大的椎旁肌,對維持腰椎穩定以及控制脊柱活動起到關鍵性的作用。近年來越來越多國內外學者聚焦于多裂肌的康復和功能訓練[1-2],以期通過促進多裂肌損傷后修復改善腰背功能障礙。骨骼肌損傷后修復主要依賴骨骼肌干細胞-肌衛星細胞(Muscle Satellite Cells,MSCs)的增殖與分化。研究證明[3-4]MSCs的增殖與分化過程有多種細胞因子及蛋白的參與,其中叉頭蛋白(Foxo1)、肌肉生長抑制素(Myostatin)及成肌分化因子(Myod)均與MSCs的增殖密切相關。……
