柬埔寨柯拉斯那沉香的化學(xué)成分研究

摘 要：該文采用 ODS、硅膠、Sephadex LH-20 等柱色譜技術(shù)，對柬埔寨野生柯拉斯那沉香（Aquilaria crassna）進(jìn)行了研究。結(jié)果表明：從柬埔寨柯拉斯那所產(chǎn)沉香的乙醇提取物中進(jìn)行分離共得到了10個化合物，包括一對對映異構(gòu)體（9a/9b）。經(jīng)波譜解析分別鑒定為6-甲氧基-2- [2-（3-羥基-4-甲氧基苯）乙基]色酮（1）、6-甲氧基-2- [2-（3-甲氧基-4-羥基苯）乙基]色酮（2）、6，7-二甲氧基-2-（2-苯乙基）色酮（3）、6-羥基-2-（2-苯乙基）色酮（4）、6-羥基-2- [2-（4-甲氧基苯）乙基]色酮（5）、8-氯-6-羥基-2- [2-（3-羥基-4-甲氧基苯）乙基]色酮（6）、8-氯-6-羥基-2- [2-（4-甲氧基苯）乙基]色酮（7）、oxidoagarochromone B（8）、4′-demethoxyaqusisnenoned（9）。其中，化合物6、7和9均為首次從柯拉斯那沉香中分離得到。活性測試結(jié)果顯示，化合物1和2對乙酰膽堿脂酶具有一定的抑制活性，化合物2對人慢性髓原白血病細(xì)胞K562具有較弱的抑制作用。

Abstract：Ten compounds， including one pair of enantiomers （9a/9b）， were isolated from the ethanol extract of agarwood originating from Aquilaria crassna in Cambodia by comprehensive chromatographic techniques including ODS， silica gel and Sephadex LH-20. On the basis of spectroscopicdata， they were identified as 6-methoxy-2- [2-（3-hydroxy-4-methoxyphenyl） ethyl] chromone （1）， 6-methoxy-2- [2-（3-methoxy-4-hydroxyphenyl） ethyl] chromone （2）， 6，7-dimethoxy-2-（2-phenylethyl） chromone （3）， 6-hydroxy-2-（2-phenylethyl） chromone （4）， 6-hydroxy-2- [2-（4-methoxyphenyl） ethyl ] chromone（5）， 8-chloro-6-hydroxy-2- [2- （3-hydroxy-4-methoxyphenyl） ethyl]chromone （6）， 8-chloro-6-hydroxy-2- [2- （4-methoxyphenyl） ethyl] chromone （7）， oxidoagarochromone B （8）， 4′-demethoxyaqusisnenoned （9）. Compounds 6， 7 and 9 were isolated from agarwood of A. crassna for the first time. Among those， compounds 1 and 2 showed inhibitory activities against acetylcholinesterase. Compound 2 had weak inhibitory effect on K562 cell lines.

沉香為瑞香科（Thymelaeaceae）沉香屬（Aquilaria）或擬沉香屬（Gyrinops）植物含有樹脂的干燥芯材（中國藥典委員會，2015）。沉香是傳統(tǒng)的名貴中藥和香料，其味辛、苦，性微溫，具有行氣止痛、溫中止嘔、納氣平喘等功效，主要用于治療胸腹脹悶疼痛、胃寒嘔吐呃逆、胃虛氣逆喘急等（中國藥典委員會，2015）。現(xiàn)代藥理學(xué)研究發(fā)現(xiàn)，沉香具有鎮(zhèn)痛、鎮(zhèn)靜、抗炎、抗氧化、抗腫瘤、細(xì)胞毒、抑菌、降糖、抗抑郁等藥理作用（戴好富，2017;楊洋等，2016;康科星等，2017）。目前報道的沉香屬植物約有23種，主要分布于中國、泰國、印度、緬甸、老撾、柬埔寨、馬來西亞、印度尼西亞等東南亞國家（戴好富和梅文莉，2015）。國內(nèi)外學(xué)者對沉……
