北京油雞和海蘭褐殼蛋雞種公雞精液品質(zhì)比較及C2CD2和RYR2基因的差異表達(dá)分析

摘要 [目的]比較北京油雞和海蘭褐殼蛋雞種公雞的精液品質(zhì)和性腺發(fā)育情況,并分析2品種間C2CD2和RYR2基因的表達(dá)差異。[方法]試驗(yàn)選取33周齡北京油雞和海蘭褐殼蛋雞種公雞各32只,從35周齡開始進(jìn)行為期10周的精液品質(zhì)檢測(cè)。從2品種的每個(gè)重復(fù)中隨機(jī)挑選1只種公雞進(jìn)行屠宰,記錄其活體重、左右睪丸重以及雞冠重并分析睪丸指數(shù)等,應(yīng)用熒光定量PCR方法檢測(cè)C2CD2和RYR2基因在2品種睪丸組織中的表達(dá)水平。[結(jié)果]海蘭褐殼蛋雞種公雞的精子密度、精子總數(shù)和性腺發(fā)育情況均優(yōu)于北京油雞。C2CD2基因在北京油雞中的表達(dá)量高于海蘭褐殼蛋雞,而RYR2基因在北京油雞中的表達(dá)量顯著低于海蘭褐殼蛋雞(P<0.05)。[結(jié)論]海蘭褐殼蛋雞的精子密度、精子總數(shù)和性腺發(fā)育情況均優(yōu)于北京油雞,RYR2基因可作為提高雞精子總數(shù)的候選基因。

Abstract [Objective] To compare the semen quality, gonadal development status of cock bred between BeijingYou chicken and Hyline brown laying hens, and analyze the differential expression of C2CD2 and RYR2 genes between the two breeds. [Method] 32 33weekold cock breeds of BeijingYou chicken and Hyline brown laying hens were selected. The semen quality testing was carried out for 10 weeks from 35weekage. One cock bred was randomly selected for slaughter from each repeat of two breeds, and the live weight, left and right testis weight and comb weight were recorded and the testicle index was analyzed. The expression level of C2CD2 and RYR2 genes in testicular tissue of two breeds was detected by Realtime PCR.[Result] The sperm density, total sperm count and gonad development of Hyline Brown laying hens were better than those of BeijingYou chicken. The expression amount of C2CD2 gene in BeijingYou chicken was higher than that of Hyline Brown laying hens, and the expression amount of RYR2 gene in BeijingYou chicken was significantly lower than that of Hyline Brown laying hens(P<0.05). [Conclusion] The sperm density, total sperm count and gonad development of Hyline Brown laying hens were better than that of BeijingYou chicken. RYR2 genes can be used as candidate genes for total sperm count of chicken.

隨著現(xiàn)代集約化高效養(yǎng)禽業(yè)的發(fā)展,人工授精技術(shù)在實(shí)際生產(chǎn)中已經(jīng)普遍應(yīng)用。在大規(guī)模種雞生產(chǎn)中,種蛋受精率會(huì)影響種蛋和雛雞的質(zhì)量,也會(huì)影響孵化率,從而影響育成期的產(chǎn)蛋性能,最終影響蛋雞養(yǎng)殖場(chǎng)的經(jīng)濟(jì)效益[1-3]。精液品質(zhì)是決定種蛋受精率的重要因素之一。北京油雞是我國(guó)的優(yōu)良地方品種,肉蛋品質(zhì)兼優(yōu),但產(chǎn)蛋量較低。海蘭褐殼蛋雞是美國(guó)海蘭國(guó)際公司培育的四系配套優(yōu)良蛋雞品種,飼料報(bào)酬、產(chǎn)蛋量和成活率均較高[4-5]。筆者選取這2個(gè)有代表性的品種進(jìn)行精液品質(zhì)和性腺發(fā)育情況的比較。

C2CD2(C2 domaincontaining 2)和RYR2(ryanodine receptor 2)基因均在維持細(xì)胞內(nèi)鈣穩(wěn)態(tài)過(guò)程中發(fā)揮生物學(xué)作用,而細(xì)胞內(nèi)鈣穩(wěn)態(tài)以及Ca2+濃度在生精細(xì)胞分化成熟、精子頂體反應(yīng)和獲能過(guò)程中均發(fā)揮著重要的作用。因此,研究C2CD2基因和RYR2基因在北京油雞和海蘭褐殼蛋雞中的表達(dá)差異,可為雞精液品質(zhì)的候選基因挖掘奠定理論基礎(chǔ)。

1 材料與方法

1.1 試驗(yàn)動(dòng)物

試驗(yàn)選取33周齡、體況一致的北京油雞和海蘭褐殼蛋雞種公雞各32只。將每個(gè)品種進(jìn)行隨機(jī)分組,每個(gè)品種4個(gè)重復(fù),每個(gè)重復(fù)8只雞。試驗(yàn)雞飼養(yǎng)與管理?xiàng)l件相同。

4 結(jié)論

海蘭褐殼蛋雞種公雞的精子密度、精子總數(shù)和性腺發(fā)育情況均優(yōu)于北京油雞,RYR2基因可作為提高雞精子總數(shù)的候選基因。
