【摘要】 目的:利用響應面Box-Behnken設計優化超聲提取桑椹多酚工藝條件。方法:選擇乙醇濃度、桑椹粉碎度、料液比、提取時間4個因素進行單因素考察,在此基礎上,用響應面Box-Behnken設計對影響較大的單因素進行3因素3水平響應面分析,以確定最佳提取條件。結果:桑椹多酚類化合物的最佳提取工藝為乙醇濃度為44.6%、桑椹粉碎度為120目、料液比為1∶34(g/ml)。結論:響應面Box-Behnken設計能優化超聲提取桑椹多酚提取條件,提高桑椹多酚提取得率。

Abstract:Objective To optimize ultrasonic extraction technology process conditions of polyphenol from Mori Fructus by the response surface method. Method Base on ethanol concentration, comminution degree, the liquid-solid ratio, ultrasonic time of single factor experiment, the BoxBehnken design for 3 star factor 3 level response surface methodology was applied, to ascertain the best extraction condition. Results The ethanol concentration of 44.6%, comminution degree of 120 section and liquid-solid ratio of 34.1∶1 were selected as the optimum conditions. Conclusion Box-Behnken design of response surface methodology was applied to optimize ultrasonic extraction technology process conditions of polyphenol from Mori Fructus

桑椹Mori Fructus為桑科植物桑(Morus alba L.)的果實,為藥食兩用之佳品,具有滋陰補血、生津潤燥的功效,用于肝腎陰虛、眩暈耳鳴、心悸失眠、須發早白、津傷口渴、內熱消渴、腸燥便秘[1]。《本草綱目》記載“椹有烏、白二種”。《四時月令》云“四月宜飲桑椹酒,能理百種風熱。”《本草新編》有云“四月采桑椹數斗,飯鍋蒸熟曬干即可為末”。多酚廣泛存在于植物體中,也是桑椹中的主要成分,其主要包括黃酮類、棓酸類及黃烷醇類等,具有抗氧化、抗菌、防癌抗癌等作用[2-4]。超聲廣泛用于有效成分提取,其可加速成分進入溶劑,保持被浸提的生物活性物質在短時間內保持不變,極大提高提取效率[5]。Box-Behnken設計是用來評價因素指標和因素間非線性關系的一種試驗設計方法,具有試驗次數相對較少,評價準確的優點,廣泛用于中藥有效成分的提取條件優化。本文在單因素實驗的基礎上,運用響應面Box-Behnken設計優化超聲提取桑椹多酚條件,為今后桑椹多酚資源更好地利用墊定了基礎。……
