Abstracts: The effects of Vitamin B1 on the growth and development and physiological and biochemical indexes of Dioscorea bulbifera L. plantlets were studied. The results showed that when vitamin B1 concentration was 0.25 mg/L, the number of bud, the bud length, the root number, the root length, the leaf number, the leaf length and the plant height could increase the number of Dioscorea bulbifera L. plantlets. When the concentration of vitamin B1 increased, the morphological indexes of Dioscorea bulbifera showed a decreasing trend. The results of physiological and biochemical indexes determination showed that when vitamin B1 concentration was 0.25 mg/L, the total chlorophyll content and the activity of POD and SOD increased, but the contents of malonaldehyde(MAD) and proline, the membrane permeability decreased. When the concentration of vitamin B1 increased, the total chlorophyll content and the activity of POD and SOD decreased, while the contents of MAD and proline, the membrane permeability increased. Under the conditions of containing vitamin B1, the content of proline of plantlets was significantly positively correlatied with the content of MDA and themembrane permeability, but there was significant negative correlation between the total content of chlorophyll and the activity of POD and SOD. The changes of content of proline can be used as a physiological index to determine the growth and development inhibition degree of D. bulbifera L. plantlets..

黃獨(Dioscorea bulbifera L.)為薯蕷科薯蕷屬植物,是世界廣布種,在中國主要分布于浙江、江西、福建等地[1]。

1.1 材料

黃獨試管苗,由上饒師范學院生命科學學院植物組織培養室提供。主要使用儀器有三角瓶、高壓滅菌鍋,超凈工作臺等。

1.2 方法

將黃獨帶芽莖段(0.5~1.5 cm)接種至100 mL三角瓶中進行單因子試驗。每個三角瓶均加入液體培養基(MS+KT 2.0 mg/L+NAA 0.1 mg/L)40 mL。試驗處理分成5組,各組的維生素B1濃度依次為0、0.25、0.50、1.00、2.00 mg/L。以上培養基pH調節為5.8~6.0,配制后在高壓滅菌鍋中滅菌20 min(121 ℃、0.1 MPa),接種都在超凈工作臺上進行。接種后的培養條件如下:光照時間14 h/d,光照度1 000~1 600 lx,溫度25 ℃,濕度70%~80%。每隔7 d觀察并記錄黃獨帶芽莖段新生芽數、根數、葉片數、芽長、根長、葉片長和株高。培養第50天對5組試管苗的生理生化指標進行測定,總葉綠素(葉綠素a和葉綠素b)含量的測定采用丙酮比色法[6],脯氨酸含量的測定采用酸性茚三酮比色法[6],過氧化物酶(POD)活性的測定采用愈創木酚法[6],超氧化物歧化酶(SOD)活性的測定采用NBT光還原法[6],丙二醛(MDA)含量的測定采用硫代巴比妥酸法[6],細胞膜透性的測定采用相對電導率法[6]。試驗重復3次,所有統計數據均表示為平均值±標準差。各組試驗數據用SPSS 19.0統計軟件進行One-Way ANOVA分析,再進行LSD法檢驗。
