細(xì)菌鞭毛染色培養(yǎng)條件的研究

[摘要] 目的 比較細(xì)菌在不同培養(yǎng)基和不同菌齡條件下鞭毛的染色效果，探索細(xì)菌鞭毛染色形態(tài)鑒定理想的培育基和培育時(shí)間。 方法 ①將普通變形桿菌分別接種在肉湯培養(yǎng)基、瓊脂培養(yǎng)基、血瓊脂培養(yǎng)基3種不同培養(yǎng)基內(nèi)，37℃培養(yǎng)18 h后，染色鏡檢，選擇取出染色效果最好的理想培養(yǎng)基；②將普通變形桿菌接種到理想培養(yǎng)基上分別培育8、12、18、24 h不同時(shí)間后，染色鏡檢，選擇出細(xì)菌鞭毛染色效果最好的培育時(shí)間。 結(jié)果 ①血瓊脂中培養(yǎng)的細(xì)菌鞭毛結(jié)構(gòu)清晰、形態(tài)完整、背景干凈，鏡檢效果最好；②12～18 h菌齡的細(xì)菌菌體中等大小，鞭毛清晰完整，鏡檢效果好。 結(jié)論 血瓊脂是鞭毛染色教學(xué)和形態(tài)鑒定理想的培養(yǎng)基；12～18 h是細(xì)菌在血瓊脂培養(yǎng)基中的最佳生長(zhǎng)時(shí)間。

The study of culture conditions in bacterial flagella staining

[Abstract] Objective To compare the bacterial flagella staining effect in different medium and different age conditions， and to explore the best medium and incubation time for bacterial flagella staining teaching and morphological identification. Methods ①The Proteus vulgaris were inoculated in broth medium， agar medium， and blood agar medium 3 different media， cultured for 18 hours based on 37℃， then stained and checked with the microscopic， and the best medium was chosen. ②The Proteus vulgaris were inoculated into the ideal medium culture for 8， 12， 18， 24 h respectively， then stained and checked with microscopic， and the best time of cultivation in flagella staining effect was selected. Results ①The bacterial growth on blood agar had good results， with complete structure， clear morphological and clean background. ②12-18 hours age had the best effect with clear and complete flagella. Conclusion The blood agar is the best medium for bacterial flagella staining teaching and morphological identification. Meanwhile， the best time of bacteria in culture is 12-18 hours.

細(xì)菌鞭毛是指存在于細(xì)菌細(xì)胞壁外的呈波浪彎曲的細(xì)長(zhǎng)絲狀物，是細(xì)菌的運(yùn)動(dòng)器官。細(xì)菌鞭毛在不同細(xì)菌中的著生位置和數(shù)量是不一樣的，是細(xì)菌種類鑒定和分類的重要依據(jù)之一[1-4]。細(xì)菌的鞭毛非常纖細(xì)，長(zhǎng)5～20 μm，直徑僅10～20 nm，小于光學(xué)顯微鏡的0.2 μm的分辨率，因此，鞭毛需經(jīng)過(guò)增粗直徑后再染色才能在光學(xué)顯微鏡下觀察[5]。鞭毛的染色效果與細(xì)菌的培養(yǎng)條件和培養(yǎng)基的類型密切相關(guān)，如劉少有等[6]發(fā)現(xiàn)嗜肺軍團(tuán)菌在BCYE培養(yǎng)基上生長(zhǎng)的鞭毛較NYE培養(yǎng)基中數(shù)量多、生長(zhǎng)好；……
