摘要:從桂林市部分公園采集了80份土樣,分離得到11株蘇云金芽孢桿菌,并對(duì)其殺蟲(chóng)晶體蛋白的形態(tài)及表達(dá)情況進(jìn)行了研究。通過(guò)光學(xué)顯微鏡觀察,發(fā)現(xiàn)這些菌株產(chǎn)生的伴孢晶體有菱形、球形、不規(guī)則形等。利用SDS-PAGE對(duì)蛋白進(jìn)行分析,結(jié)果表明9株蘇云金芽孢桿菌表達(dá)的蛋白大小約為135 ku,5株表達(dá)蛋白大小約65 ku,1株表達(dá)蛋白大小約90 ku,1株表達(dá)蛋白大小約45 ku,5株同時(shí)表達(dá)約135 ku和65 ku兩種蛋白。產(chǎn)小菱形晶體的菌株7-4可能對(duì)美國(guó)白蛾有高毒力,值得進(jìn)一步深入研究。

Isolation of Bacillus thuringiensis from parks in Guilin

Abstract: 11 Bacillus thuringiensis(Bt) isolates were obtained from 80 soil samples collected from some parks in Guilin,and the morphology and expression of insecticidal crystal proteins from these strains were futher studied. The optical microscopic observation showed that there were bipyramidal, spherical, and irregular parasporal crystals in the 11 Bt isolates. Analysis of proteins by SDS-PAGE indicated that 9 Bt isolates could express proteins with 135 ku molecular mass, 5 strains with 65 ku, 1 strain with 90 ku, 1 strain with 45 ku, and 2 isolates expressed both 130 ku and 60 ku proteins. Strain 7-4 producing small bipyramidal crystal protein may have high toxcity to Hyphantria cunea(Drury), which deserved to be further studied.

蘇云金芽孢桿菌(Bacillus thuringiensis,Bt)是一種在自然界中廣泛分布的革蘭氏陽(yáng)性芽孢桿菌,是目前世界上用途最廣、應(yīng)用最成功、產(chǎn)量最大的微生物殺蟲(chóng)劑[1],可作為化學(xué)農(nóng)藥的替代品[2,3],已成為生物農(nóng)藥研究的重點(diǎn)和熱點(diǎn)。Bt在芽孢形成過(guò)程中,能在菌體的一端或兩端產(chǎn)生一個(gè)或多個(gè)形狀一致或不同的伴孢晶體(Parasporal crystal)[1,2],又稱(chēng)殺蟲(chóng)晶體蛋白(Inseticidal crystal proteins,ICPs)或δ-內(nèi)毒素(Delta-endotoxin)[4,5],由cry或cyt基因編碼,是Bt殺蟲(chóng)活性的主要來(lái)源[6]。研究表明,ICPs具有高效、廣譜的殺蟲(chóng)作用,對(duì)人畜及非目標(biāo)昆蟲(chóng)無(wú)害,對(duì)鱗翅目、雙翅目、鞘翅目、膜翅目、同翅目、直翅目、食毛目以及吸蟲(chóng)、鞭毛蟲(chóng)、線蟲(chóng)、螨類(lèi)和原生動(dòng)物等200多種害蟲(chóng)具有毒力作用[5,7-9],且具有抗癌作用[10]。……
