HPLC法測定菊明降壓片中蒙花苷的含量

【摘 要】 目的：建立菊明降壓片中蒙花苷的含量測定方法。方法：色譜柱：Hypersil ODS2（4.6mm×150mm，5μm）；流動相：乙腈-0.5%磷酸溶液（30：70）；檢測波長：334nm；流速：0.6ml/min；進樣20μl。結果：蒙花苷在3-30μg/ml范圍內，峰面積與其濃度呈良好線性關系（r=0.9994），平均回收率為93.61%，RSD為1.15%（n=4）。結論：該方法簡便、準確、重復性好，可用于測定菊明降壓片中蒙花苷的含量。

Determination the Content of Buddleoside in Juming Jiangya

【Abstract】 Objective To establish a HPLC method for analysis the content of buddleoside in Juming Jiangya Tablet by HPLC . Method The separation was performed on C18 Column （250mm×4.6mm） with the mobile phase consisted of acetonitrile-0.5% Phosphoric acid （30：70，V/V）.The flow rate was 0.6ml/min，the injection volume was 20μl， and the detection wave length was 334nm and the column temperature was room temperature. Result Linearity of buddleoside was good at 3-30μg/ml（r=0.9994）.The average recovery of buddleoside was 93.61% with RSD=1.15%（n=4）.Conclusion The method is simple，accurate and repeatable，which can be applied in determination the content of buddleoside in Juming Jiangya tablet.

菊明降壓片，是野菊花和決明子以4：1的比例經提取制得的復方制劑。性狀為棕褐色，味苦、澀。菊明降壓片對原發性高血壓，慢性腎炎性高血壓等的治療有很好的療效。

野菊花為菊科多年生草本植物野菊等的頭狀花序，全國各地均產[1]。野菊花，味苦、辛，性涼，清熱解毒，疏風平肝，用于疔瘡、癰腫、丹毒，風熱感冒、咽喉腫痛、高血壓眩暈、頭痛等病癥。野菊花含有揮發油、黃酮、綠原酸等多種有效化學成分，其中蒙花苷是野菊花中重要的黃酮類化合物，具有明顯的抗菌活性等多種作用[2，3]。蒙花苷（Buddleoside ，別名Linarin）是從菊科植物野菊Chrysanthemum indicum L.的頭狀花序中提取的化合物，為白色粉末，分子式為C28H32O14，分子量為592.5468，屬黃酮類藥物，具有抗衰老、抗缺氧、抗不良刺激、抗疲勞、抑制血糖升高和對機體雙向調節作用，對學習記憶也有影響，一般將其作為野菊花的指標成分。

菊明降壓片收載在《衛生部藥品標準》中，其質量標準中沒有含量測定項[4]，不利于產品質量的控制，因此，建立測定菊明降壓片含量的方法，對控制該品種質量有重要意義。本實驗擬以蒙花苷為指標成分[5-7]，建立HPLC法測定菊明降壓片中的蒙花苷的含量的方法，以實現該品種的含量控制，方法未見報道。……
