



[摘要] 目的 應用單細胞凝膠電泳法和CB微核+3AB指數實驗檢測苯作業工人的DNA修復能力及個體敏感性,探討DNA修復能力及個體敏感性與慢性苯中毒的關系。方法 采用固定群體隨機抽樣,選取慢性苯中毒工人及其對照,檢測彗星尾長差和CB微核+3AB指數;應用統計學方法對相關指標進行分析。 結果 苯中毒組和對照組的彗星尾長差沒有顯著性差異(P>0.05),而兩組間的CB微核+3AB指數有顯著性差異(P<0.05)。苯作業工人發生慢性苯中毒危險性與彗星尾長差之間趨勢關系沒有顯著性(?字2趨勢=2.588,P=0.108)但與3AB指數之間存在顯著趨勢關系(?字2趨勢=9.100,P=0.003)。結論 苯作業工人在苯接觸相近的情況下,個體易感性是苯作業工人發生苯中毒與否的決定性因素。DNA損傷修復能力低下即個體易感性高的個體接觸苯時發生苯中毒的危險性增加。

Risk analysis of chronic benzene poisoning and individual susceptibility

[Abstract] Objective We applied Single Cell Gel Electrophoresis assay and Cytokinesis-block micronucleus and 3AB index method to measure individual susceptibility, and detected relationship of chronic benzene poisoning and individual susceptibility.Methods We investigated comettail length difference and CBMN+3AB index. And the related factors were analyzed by statistical methods. Results There is no significant difference of comettail difference between benzene poisonings and their controls(P>0.05); While the difference of CBMN+3AB index between them is significant(P<0.05). The trend of comettail difference and benzene poisoning is not significant(?字2趨勢=2.588,P=0.108), but 3AB index and benzene poisoning is significant (?字2趨勢=9.100, P=0.003). Conclusion In the similar benzene exposure, individual genetic susceptibility of benzene poisonings is higher because of lower DNA repair ability.

正常細胞擁有完整的DNA修復系統,從而防止DNA永久損傷的發生并維持遺傳物質的完整性。DNA修復是一個非常復雜的過程,涉及許多酶和蛋白質的參與,一旦有關基因發生突變,就會導致DNA修復能力降低。在某些染色體不穩定綜合征患者的體細胞內已經發現了有關DNA修復基因突變和表達異常,這些結果為揭示DNA修復能力低下的分子機制和易感人群的篩查奠定了基礎,DNA修復能力的高低即個體易感性是判別損害預后及對損害程度評估的重要依據。……
