[摘要] 目的 探討大豆異黃酮對非酒精性脂肪肝病模型大鼠抗氧化能力的影響。方法 將50只SD大鼠隨機分成5組:正常組,模型組,大豆異黃酮低劑量組,大豆異黃酮中劑量組,大豆異黃酮高劑量組;除正常組外,其余各組以高脂飲食誘導(dǎo)建立大鼠脂肪肝模型,測定肝組織中MDA、SOD和T-AOC水平,對肝臟進行病理學(xué)檢查,觀察大豆異黃酮對該模型的影響。結(jié)果 大豆異黃酮可降低脂肪肝大鼠MDA,提高SOD和T-AOC水平,并能明顯改善模型動物肝組織的病理損傷。結(jié)論 大豆異黃酮提高非酒精性脂肪肝病大鼠的抗氧化能力,利于脂肪肝的治療。

Efects of Soybean Isoflavones on the Antioxidant Activity of Fatty Liver Model in Rats

[Abstract] Objective To evaluate the protective efects of Soybean Isoflavones on the antioxidant activity of nonalcoholic fatty liver diseases in rats. Methods 50 SD rats were randomly divided into 5 groups: nomal control group, model group, low dose of Soybean Isoflavones treatment group, intermediate dose of Soybean Isoflavones treatment group and high dose of Soybean Isoflavones treatment group. The fatty liver model of rats were induced by oral administration with high fat diet. Live tissue in animal was extracted to tested the MDA、SOD and T-AOC, Histopathologic examination of the liver tissues was also performed. Results Soybean Isoflavones was found to reduce the level of MDA and increase SOD and T-AOC levels in the liver tissues of fatty liver model, Soybean Isoflavones also markedly ameliorated the pathological injury in the liver tissue. Conclusion Soybean Isoflavones can exert a significant protective effect on nonalcoholic fatty liver diseases in rats by icrease their ability of antioxidant activity.

近年來非酒精性脂肪肝(nonalcoholic fatty liver diseases,NAFLD)發(fā)病率呈逐年上升的趨勢。NAFLD的發(fā)病機制較復(fù)雜,有廣泛的因素參與,迄今尚未完全闡明,其中氧化應(yīng)激相關(guān)性損害是其重要的病理環(huán)節(jié)。大豆異黃酮(Soybean Isoflavones,SI)具有很強的抗氧化活性,如清除超氧陰離子、羥自由基和過氧自由基或單線態(tài)氧[1]。本研究旨在觀察大豆異黃酮對大鼠高脂飲食性脂肪肝模型的抗氧化作用。
1材料與方法
1.1實驗動物
成年SD大鼠,雌雄各半,體重180-200g 由延邊大學(xué)醫(yī)學(xué)部實驗動物中心提供。
1.2試劑和儀器
大豆異黃酮系天然提取物,由成都牧甫生物科技有公司提供。超氧化物歧化酶(SOD)、丙二醛(MDA)、總抗氧化能力(T-AOC)試劑盒購于南京建成試劑公司。752型紫外光柵分光光度計為上海第三分析儀器廠產(chǎn)品;236HK臺式冷凍離心機為德國HERMLE產(chǎn)品;德國Sartarius的BP211D電子天平。……
